Urine Testing for Sugar and Ketone

Urine Testing is a test to determine the amount of sugar and ketone in the urine.


Materials:

1. Fresh Urine from patients in a clean container.
2. Clinitest reagent tablet as indicated, acetest tablet as indicated, Benedict's solution as indicated.
3. Test tube, clean and dry
4. Index card
5. Medicine Dropper
6. Alcohol lamp


Procedure:


Urine Collection



1. Have the patient empty his bladder 1/2- 1 hour before the actual collection of urine for testing. Discard this initial urine.

2. Let the patient void again 30 min after.

3. Collect a small amount of urine (20cc) in a clean container or specimen bottle.


Test for Urine Sugar

a. Proceed for urine collection.

b. Holding a dropper upright, put 5 drops of urine into a clean, dry test tube.

c. Rinse dropper. Using the same dropper, add 10 drops of water into the test tube with urine.

d. Drop 1 clinitest tablet into the test tube.

e. Allow reaction to take place until it stops.

f. Wait for 15 seconds. Shake gently. Compare color results in the test tube with chart.

     * blue- negative result
     * orange- highly positive result
     * dark greenish-brown proceeded by rapid change in color from green to orange- indicates urine glucose level above 2%


Acetone (Ketone Test)

a. Place acetest tablet on a piece of paper towel.

b. Place 1 drop of urine in a acetest tablet with dropper in upright position.

c. Wait for a minute and compare color of tablet with chart.

     * negative result- tablet color unchanged or turns cream colored from wetting
     * positive result- tablet color will change from lavender to deep purple depending of amount of ketone bodies present.


Benedict's Test

a. Follow urine collection.

b. In a clear, dry test tube, place 5cc of Benedict's solution.

c. Add 8-10 drops of urine to the solution and place test tube with mixture over a direct flame or in a water bath to boil for 5 minutes.

d. Compare color with index card

     * blue- negative
     * orange- +
     * yellow- ++
     * yellow orange- +++
     * red- ++++
